Abstract
Latar belakang: Autism Spectrum Disorder (ASD) merupakan gangguan perkembangan neurologis yang kompleks. Anak dengan ASD rentan mengalami infeksi saluran kemih (ISK) karena adanya komorbiditas perilaku seperti kebiasaan menahan buang air kecil. Pemeriksaan urin yang tertunda dapat mempengaruhi akurasi hasil, khususnya dalam pemeriksaan total bakteri. Tujuan: Penelitian ini dilakukan untuk mengetahui hasil pemeriksaan total bakteri pada urin anak ASD berdasarkan penyimpanan sampel pada suhu ruang dengan perlakuan lama waktu penyimpanan sampel 0 jam, 1,5 jam, 3 jam, 4,5 jam, dan 6 jam. Metode: Jenis penelitian ini adalah eksperimental laboratorium dengan desain cross-sectional. Sampel urin anak ASD diambil secara midstream, lalu dilakukan pemeriksaan total bakteri menggunakan teknik kultur kuantitatif (teknik mayo). Hasil uji berupa jumlah koloni yang tumbuh pada media MacConkey Agar dikalikan dengan 102 CFU/mL. Data dianalisis menggunakan uji statistik non-parametrik menggunakan uji spearman sesuai distribusi data. Hasil: Berdasarkan hasil penelitian pada pemeriksaan segera (0 Jam) didapatkan rata-rata jumlah koloni sebesar 0,2 x 102 CFU/mL, setelah dilakukan penyimpanan pada suhu ruang dengan waktu penundaan pemeriksaan 1,5 jam didapatkan rata-rata jumlah koloni sebesar 0,23 x 102 CFU/mL, pada penundaan pemeriksaan 3 jam sebesar 0,1 x 102 CFU/mL, pada penundaan pemeriksaan 4,5 jam sebesar 0,2 x 102 CFU/mL, dan pada penundaan pemeriksaan 6 jam sebesar 5,76 x 102 CFU/mL. Sedangkan hasil uji spearman menunjukkan nilai signifikansi (p) 0.000 dengan arah korelasi positif, yang berarti lama penundaan pemeriksaan berpengaruh terhadap jumlah koloni bakteri

Background: Autism Spectrum Disorder (ASD) is a complex neurodevelopmental disorder. Children with ASD are more susceptible to urinary tract infections (UTIs) due to behavioral comorbidities, such as the habit of delaying urination. Delayed urine examination may affect the accuracy of laboratory results, particularly the determination of urinary bacterial counts. Objective: This study aimed to determine the total urinary bacterial counts in urine samples collected from children with ASD following storage at room temperature for 0, 1.5, 3, 4.5, and 6 hours before examination. Methods: This laboratory experimental study employed a cross- sectional design. Midstream urine samples were collected from children with ASD and analyzed for total urinary bacterial counts using the quantitative urine culture method (Mayo technique). Bacterial colonies grown on MacConkey agar were enumerated and expressed as × 10² CFU/mL. Data were analyzed using Spearman's rank correlation test, a non-parametric statistical test selected according to the data distribution. Results: Immediate examination (0 hours) yielded a mean bacterial count of 0.2 × 10² CFU/mL. After storage at room temperature, the mean bacterial counts were 0.23 × 10² CFU/mL after 1.5 hours, 0.1 × 10² CFU/mL after 3 hours, 0.2 × 10² CFU/mL after 4.5 hours, and 5.76 × 10² CFU/mL after 6 hours. Spearman's rank correlation analysis demonstrated a statistically significant positive correlation (p < 0.001), indicating that storage time before examination significantly affected bacterial colony counts.
