Abstract

Infeksi Saluran Kemih (ISK) merupakan salah satu infeksi yang sering terjadi dan sebagian besar disebabkan oleh Escherichia coli. Metode kultur urine sebagai baku emas memiliki keterbatasan karena membutuhkan waktu yang relatif lama. Diperlukan metode diagnosis yang lebih cepat dan sensitif, salah satunya menggunakan teknik PCR. Adapun tujuan penelitian ini untuk mendeteksi DNA Escherichia coli pada sampel urine pagi pasien suspek ISK menggunakan metode PCR. Jenis penelitian yang digunakan pada penelitian ini adalah deskriptif laboratorik dengan pendekatan cross sectional, sedangkan untuk Teknik sampling yang digunakan adalah total sampling. Hasil penelitian menunjukkan bahwa seluruh sampel berhasil diisolasi namun tidak seluruh sampel mengandung DNA target Escherichia coli, dengan kadar dan nilai kemurnian yang bervariasi. Konsentrasi dan kemurnian DNA mencerminkan kualitas dan jumlah total, sedangkan terbentuknya pita DNA target pada hasil visualisasi elektroforesis menjadi indikator utama keberhasilan isolasi DNA E.coli. Berdasarkan hasil visualisasi elektroforesis PCR, diperoleh sampel positif terdapat DNA E.coli sebesar (62,5%). Sementara itu, diperoleh sampel negatif sebesar (37,5%). Metode PCR mampu mendeteksi DNA target Escherichia coli pada sampel urine pagi pasien suspek ISK secara spesifik didukung penerapan quality control pada setiap tahapan pemeriksaan.

Urinary tract infections (UTIs) are among the most common infections and are primarily caused by Escherichia coli. Urine culture, considered the gold standard, has limitations because it takes a relatively long time. A faster and more sensitive diagnostic method is needed, one of which is the PCR technique. The objective of this study was to detect Escherichia coli DNA in morning urine samples from patients with suspected UTIs using the PCR method. This study employed a descriptive laboratory design with a cross-sectional approach, and total sampling was used. The results showed that DNA was successfully isolated from all samples; however, not all samples contained the target Escherichia coli DNA, with varying concentrations and purity levels. The DNA concentration and purity reflect the quality and total quantity, while the formation of target DNA bands in the PCR electrophoresis visualization results serves as the primary indicator of successful E. coli DNA isolation. Based on the PCR electrophoresis visualization results, 62.5% of the samples tested positive for E. coli DNA. Meanwhile, negative samples accounted for 37.5%. The PCR method was able to specifically detect Escherichia coli target DNA in morning urine samples from patients with suspected UTIs, supported by the implementation of quality control at every stage of the examination.
