Abstract
Latar Belakang: Lansia dengan komorbiditas Diabetes Melitus (DM) memiliki risiko lebih tinggi mengalami infeksi saluran kemih akibat kondisi hiperglikemia yang menyebabkan glukosuria sehingga urin menjadi media yang mendukung pertumbuhan bakteri. Penundaan pemeriksaan urin setelah pengambilan sampel pada suhu ruang dapat meningkatkan jumlah bakteri sehingga memengaruhi kualitas specimen dan hasil pemeriksaan. Tujuan: penelitian ini bertujuan untuk mengetahui hasil pemeriksaan total bakteri pada urin lansia dengan komorbiditas Diabetes Melitus (DM) menggunakan perlakuan lama waktu penyimpanan sampel pada suhu ruang. Metode: Jenis penelitian ini adalah eksperimental laboratorium dengan desain quasi experimental. Sampel urin lansia dengan komorbiditas DM diambil secara mindstream, lalu disimpan pada suhu ruang (22–27°C) dan dilakukan pemeriksaan total bakteri pada waktu 0 jam (pemeriksaan segera), penundaan 2 jam, 4 jam dan 6 jam menggunakan teknik kultur kuantitatif (Teknik mayo) untuk menghitung total bakteri dalam satuan CFU/mL. Hasil: Berdasarkan hasil penelitian pada pemeriksaan segera (0 jam) didapatkan rata-rata jumlah koloni sebesar 3,46 x 103 CFU/mL, setelah dilakukan penyimpanan pada suhu ruang dengan waktu penundaan 2 jam didapatkan rata-rata jumlah koloni sebesar 9,07 x 103 CFU/mL, pada penundaan pemeriksaan 4 jam sebesar 10,82 x 103 CFU/mL, dan pada penundaan 6 jam sebesar 12,23 x 103 CFU/mL. Uji spearman menunjukkan adanya hubungan yang signifikan antara lama waktu penundaan pemeriksaan dengan rata-rata jumlah bakteri (nilai koefisien berkisar 0.729**- 0.914** dan nilai sig. (2-tailed) 0.000).

Background: Elderly with Diabetes Mellitus (DM) comorbid have a higher risk of developing urinary tract infections due to hyperglycemia, which causes glycosuria and turns urine into a medium that supports bacterial growth. Delaying urine testing after sample collection at room temperature can increase the bacterial count, thereby affecting specimen quality and test results. Objective: This study aims to determine the results of total bacterial counts in the urine of older adults with comorbid Diabetes Mellitus (DM) by varying the duration of sample storage at room temperature. Methods: This was a laboratory experimental study using a quasi-experimental design. Urine samples from older adults with DM were collected via midstream voiding, then stored at room temperature (22–27°C). Total bacterial counts were performed at 0 hours (immediate testing), and after delays of 2 hours, 4 hours, and 6 hours using quantitative culture techniques (Mayo method) to determine total bacteria in CFU/mL. Results: Based on the study results, the immediate examination (0 hours) yielded an average colony count of 3.46 × 10³ CFU/mL; after storage at room temperature with a 2-hour delay, the average colony count was 9.07 × 10³ CFU/mL; at a 4-hour delay, the bacterial count was 10.82 × 10³ CFU/mL, and at a 6-hour delay, it was 12.23 × 10³ CFU/mL. The Spearman test showed a significant correlation between the duration of the test delay and the average bacterial count (coefficient values ranging from 0.729** to 0.914** and a 2-tailed significance value of 0.000).
