Abstract

Latar belakang penelitian ini adalah tingginya sensitivitas metode Polymerase Chain Reaction (PCR) dalam mendeteksi DNA bakteri dengan jumlah yang sangat sedikit dan waktu pemeriksaan yang lebih cepat. Pemilihan sampel darah didasarkan pada kemampuannya dalam merepresentasikan penyebaran infeksi secara sistemik, sehingga dapat digunakan untuk mengetahui kemungkinan perkembangan Infeksi Saluran Kemih (ISK) yang sebagian besar disebabkan oleh Escherichia coli menjadi bakteremia. Tujuan penelitian ini adalah untuk mengetahui keberadaan DNA E. coli pada sampel darah penderita ISK menggunakan metode PCR. Metode penelitian yang digunakan adalah deskriptif laboratorik dengan pendekatan cross sectional. Sampel penelitian berupa darah dari delapan pasien suspek ISK yang memenuhi kriteria inklusi dan eksklusi. DNA diekstraksi menggunakan QIAamp DNA Mini Kit, kemudian dilakukan pengukuran konsentrasi dan kemurnian DNA menggunakan Nanodrop-Lite Spectrophotometer. Selanjutnya, DNA diamplifikasi menggunakan primer spesifik E. coli dan divisualisasikan dengan metode agarose gel electrophoresis menggunakan Gel Documentation System (Gel Doc). Data dianalisis secara deskriptif dan disajikan dalam bentuk tabel serta diagram pie. Hasil penelitian menunjukkan bahwa lima dari delapan sampel (62,5%) menunjukkan hasil positif dengan terbentuknya pita DNA target, sedangkan tiga sampel (37,5%) menunjukkan hasil negatif. Variasi konsentrasi dan kemurnian DNA hasil isolasi ditemukan pada masing-masing sampel. Konsentrasi dan kemurnian DNA mencerminkan jumlah dan kualitas DNA total, sedangkan terbentuknya pita DNA target pada hasil agarose gel electrophoresis menjadi indikator utama keberhasilan deteksi DNA E. coli. Kesimpulan dari penelitian ini adalah metode PCR efektif digunakan untuk mendeteksi DNA Escherichia coli pada sampel darah penderita ISK.

The background of this research is the high sensitivity of the Polymerase Chain Reaction (PCR) method in detecting bacterial DNA in very small quantities and the faster examination time. The selection of blood samples is based on their ability to represent the systemic spread of infection, making them useful for determining the potential progression of Urinary Tract Infections (UTIs), which are mostly caused by Escherichia coli, into bacteremia. The aim of this study is to determine the presence of E. coli DNA in blood samples from UTI patients using PCR. The research method used is a laboratory descriptive approach with a cross-sectional design. The research samples consisted of blood from eight suspected UTI patients who met the inclusion and exclusion criteria. DNA was extracted using the QIAamp DNA Mini Kit, followed by measurement of DNA concentration and purity using the Nanodrop-Lite Spectrophotometer. Next, DNA was amplified using E. coli-specific primers and visualized using the agarose gel electrophoresis method with a Gel Documentation System (Gel Doc). Data were analyzed descriptively and presented in tables and pie charts. The research results show that five of eight samples (62.5%) tested positive for the formation of the target DNA band, while three (37.5%) tested negative. Variations in the concentration and purity of the isolated DNA were found in each sample. The concentration and purity of DNA reflect the total amount and quality of DNA. At the same time, the formation of the target DNA band in the agarose gel electrophoresis results is the main indicator of successful E. coli DNA detection. This study concludes that the PCR method is effective for detecting Escherichia coli DNA in blood samples from UTI patients.
