Abstract
Latar belakang: Kehamilan meningkatkan risiko infeksi saluran kemih (ISK) akibat pertumbuhan bakteri yang dipengaruhi perubahan fisiologis. Penundaan pemeriksaan sampel urin pada suhu ruang dapat meningkatkan jumlah bakteri sehingga memengaruhi hasil pemeriksaan. Tujuan: Mengetahui hasil pemeriksaan total bakteri pada sampel urin ibu hamil dengan perlakuan lama waktu penyimpanan 0 jam, 1,5 jam, 3 jam, dan 4,5 jam pada suhu ruang. Metode: Penelitian ini merupakan penelitian eksperimental laboratorium dengan dengan desain cross-sectional. Sampel urin ibu hamil trimester ketiga dikumpulkan dengan metode midstream, kemudian dilakukan pemeriksaan total bakteri menggunakan kultur kuantitatif (colony counting/teknik mayo). Jumlah koloni yang tumbuh pada media Mac conkey agar dikalikan 102 CFU/ml. Analisis data menggunakan uji statistik nonparametrik spearman sesuai distribusi data. Hasil: Rerata total bakteri pada perlakuan 0 jam sebesar 15,19 x 102 CFU/mL, perlakuan 1,5 sebesar 22,44 x 102 CFU/mL, perlakuan 3 jam sebesar 22,03 x 102 CFU/mL, perlakuan 4,5 jam sebesar 26,86 x 102 CFU/mL. Uji spearman menunjukkan koefisien korelasi lebih besar dari nilai signifikansi (p=0,001) yaitu 0-1,5 jam sebesar 890**, 0-3 jam sebesar 892**, 0-4,5 jam sebesar 913**, 1,5-3 jam sebesar 965**, 1,5-4,5 jam sebesar 921**, 3-4,5 jam sebesar 957**. Kesimpulan: Lama waktu penyimpanan sampel urin pada suhu ruang berpengaruh terhadap peningkatan jumlah bakteri, sehingga pemeriksaan sebaiknya dilakukan sesegera mungkin untuk memperoleh hasil yang akurat.

Background: Pregnancy increases the risk of urinary tract infections (UTIs) due to bacterial growth influenced by physiological changes. Delaying the testing of urine samples at room temperature can increase the bacterial count, thereby affecting test results. Objective: To determine the total bacterial count in urine samples from pregnant women with storage times of 0 hours, 1.5 hours, 3 hours, and 4.5 hours at room temperature. Methods: This was a laboratory-based experimental study with a cross-sectional design. Urine samples from third-trimester pregnant women were collected using the midstream method, followed by total bacterial count testing via quantitative culture (colony counting/Mayo technique). The number of colonies growing on MacConkey agar was multiplied by 10² CFU/mL. Data analysis used Spearman’s nonparametric statistical test based on the data distribution. Results: The mean total bacterial count for the 0-hour treatment was 15.19 × 10² CFU/mL, for the 1.5-hour treatment was 22.44 × 10² CFU/mL, for the 3-hour treatment was 22.03 × 10² CFU/mL, and 26.86 × 10² CFU/mL for the 4.5-hour treatment. Spearman’s test showed a correlation coefficient greater than the significance level (p = 0.001), specifically 890** for 0–1.5 hours, 0–3 hours at 892**, 0–4.5 hours at 913**, 1.5–3 hours at 965**, 1.5–4.5 hours at 921**, and 3–4.5 hours at 957**. Conclusion: The duration of urine sample storage at room temperature affects the increase in bacterial count; therefore, testing should be performed as soon as possible to obtain accurate results.
