Abstract
Latar Belakang : Infeksi Saluran Kemih (ISK) merupakan permasalah kesehatan yang memerlukan perhatian serius pada anak Autism Spectrum Disorder (ASD) akibat hambatan komunikasi dan kebiasaan menahan buang air kecil. Keterlambatan pemeriksaan urin di laboratorium rujukan seringkali menurunkan mutu hasil dan akurasi diagnosis. Tujuan: Penelitian ini bertujuan untuk mengetahui hasil pemeriksaan total bakteri pada sampel urin anak ASD dengan berbagai variasi lama penyimpanan pada suhu lemari es (2-8°C). Metode: Penelitian ini merupakan eksperimen laboratorium dengan desain cross-sectional. Sampel urin anak ASD diambil dan dilakukan perlakuan penyimpanan pada suhu lemari es dengan durasi 0 jam (kontrol), 1,5 jam, 3 jam, 4,5 jam, dan 6 jam. Teknik analisis data yang digunakan adalah uji normalitas Shapiro-Wilk yang dilanjutkan dengan uji Spearman untuk menganalisis pengaruh waktu penundaan terhadap jumlah koloni bakteri. Hasil : Berdasarkan hasil penelitian pada pemeriksaan segera (0 Jam) didapatkan rata-rata jumlah koloni sebesar 0,07 x 102 CFU/mL, setelah dilakukan penyimpanan pada suhu lemari es dengan waktu penundaan pemeriksaan 1,5 jam didapatkan rata-rata jumlah koloni sebesar 0,10 x 102 CFU/mL, pada penundaan pemeriksaan 3 jam sebesar 0,20 x 102 CFU/mL, pada penundaan pemeriksaan 4,5 jam sebesar 0,40 x 102CFU/mL, dan pada penundaan pemeriksaan 6 jam sebesar 4,30 x 102 CFU/mL. Sedangkan hasil uji spearman menunjukkan nilai signifikansi (p) 0.000 dengan arah korelasi positif, yang berarti lama penundaan pemeriksaan berpengaruh terhadap jumlah koloni bakteri.

Background: Urinary Tract Infection (UTI) is a health problem that requires serious attention in children with Autism Spectrum Disorder (ASD) due to communication barriers and urinary retention. Delays in urine testing at reference laboratories often reduce the quality of results and diagnostic accuracy. Objective: This study aimed to determine the results of total bacterial testing in urine samples from children with ASD with various storage times at refrigerator temperature (2- 8°C). Methods: This study was a laboratory experiment with a cross-sectional design. Urine samples from children with ASD were collected and stored at refrigerator temperature for 0 hour (control), 1.5 hours, 3 hours, 4.5 hours, and 6 hours. The data analysis technique used was the Shapiro-Wilk normality test followed by the Spearman test to analyze the effect of delay time on bacterial colony counts. Results: Based on the results of the study, the immediate examination (0 hour) yielded an average colony count of 0.07 x 102 CFU/mL. After storage at refrigerator temperature with a 1.5-hour delay, the average colony count was 0.10 x 102 CFU/mL. After a 3-hour delay, the count was 0.20 x 102 CFU/mL, after a 4.5- hour delay, the count was 0.40 x 102 CFU/mL, and after a 6-hour delay, the count was 4.30 x 10 CFU/mL. The Spearman test showed a significance value (p) of 0.000 with a positive correlation, indicating that the length of the delay affected the bacterial colony count.
