Abstract

Background: Exposure to stress can affect the digestive system, particularly the stomach, through increased activity of the sympathetic nervous system and the secretion of stress hormones, which can potentially cause changes in the structure of stomach tissue. Recitation of the Qur’an is a non-pharmacological therapy known to have a relaxing effect and to reduce the stress response. Objective: This study aimed to determine the effect of varying durations of murottal exposure on the histological profile of the stomach in Rattus norvegicus exposed to CO₂. Methods: This was an experimental study using a post-test only control group design. Test animals were divided into a control group and treatment groups that were exposed to the recitation of the Qur’an 1, 2, 3, 4, and 5 times per day for 10 minutes. Gastric tissue was prepared for histological examination using Hematoxylin-Eosin staining and observed under a microscope. Data analysis was performed using the Shapiro-Wilk normality test, Levene’s homogeneity test, and one-way ANOVA. Results: The observations showed that the histological appearance of the stomach was relatively similar across all groups. The structure of the stomach was still observable, with cell nuclei appearing bluish-purple and cytoplasm appearing pink. Some specimens in the groups with higher murottal exposure intensity showed a clearer tissue structure, though this was not consistently observed across all samples. The results of the statistical analysis showed a significance value of 0.006 for the normality test, 0.385 for the homogeneity test, and 0.094 for the Kruskal-Wallis test (p > 0.05), indicating that there were no significant differences among the treatment groups. Conclusion: Variations in the duration of murottal exposure did not have a significant effect on the histological appearance of the stomachs of CO₂-exposed Rattus norvegicus. Although higher exposure intensity tended to produce a clearer tissue pattern in some specimens, these differences were not statistically significant.
