Abstract

Kratom (Mitragyna speciosa Korth.) mengandung alkaloid mitraginin yang memiliki aktivitas farmakologis sebagai analgesik dan stimulan, namun berpotensi disalahgunakan sehingga diperlukan metode analisis yang tervalidasi untuk pengendalian mutu dan pengawasan. Penelitian ini bertujuan mengembangkan dan memvalidasi metode Gas Chromatography-Flame Ionization Detector (GC-FID) untuk penetapan kadar mitraginin pada tiga varietas daun kratom (merah, hijau, putih) dari Kalimantan Barat. Simplisia daun kratom yang telah distandardisasi diekstraksi dengan maserasi menggunakan etil asetat, dilanjutkan fraksinasi cair-cair asam-basa untuk pemurnian mitraginin. Analisis dilakukan menggunakan GC-FID dengan kolom HP-5 (30 m × 320 μm × 0,25 μm) pada suhu injektor 280°C, program suhu kolom 200-285°C, laju alir gas pembawa helium 1,0 mL/menit, dan rasio split 1:10. Validasi metode meliputi spesifisitas dengan TLC-densitometri (Rs ≥ 1,0), selektivitas ditunjukkan keterpisahan puncak (peak) mitraginin dari komponen matriks dan standar internal pada kromatogram, linieritas (r = 0,9993; range 5,0-100,0 mg/L), presisi (%RSD 0,7996%), dan akurasi (recovery 96,29-101,84%). Hasil penelitian menunjukkan metode GC-FID memenuhi seluruh parameter validasi sesuai ICH Q2(R2) dan AOAC. Kadar mitraginin tertinggi ditemukan pada varietas hijau (1,1321 ± 0,0194 mg/g), diikuti merah (0,6409 ± 0,0377 mg/g) dan putih (0,1510 ± 0,0031 mg/g). Analisis statistik One-Way ANOVA mengkonfirmasi perbedaan sangat signifikan antar varietas daun kratom (F =796,136; p < 0,001). Metode fraksinasi cair-cair asam-basa terbukti 31-71 kali lebih efektif dibanding ekstraksi langsung. Kesimpulannya, metode GC-FID yang telah tervalidasi ini bersifat reliabel dan aplikatif untuk analisis kuantitatif mitraginin dalam standardisasi dan pengawasan produk kratom.

Kratom (Mitragyna speciosa Korth.) contains the alkaloid mitragynine, which possesses pharmacological activities as an analgesic and stimulant; however, it has potential for abuse, necessitating a validated analytical method for quality control and surveillance. This study aimed to develop and validate a Gas Chromatography-Flame Ionization Detector (GC-FID) method for the determination of mitragynine content in three kratom leaf varieties (red, green, and white) from West Kalimantan. Standardized kratom leaf simplicia were extracted by maceration using ethyl acetate, followed by acid-base liquid-liquid fractionation for mitragynine purification. Analysis was performed using GC-FID with an HP-5 column (30 m × 320 μm × 0.25 μm) at an injector temperature of 280°C, column temperature program of 200-285°C, helium carrier gas flow rate of 1.0 mL/min, and split ratio of 1:10. Method validation included specificity by TLC-densitometry (Rs ≥ 1.0), selectivity demonstrated by separation of mitragynine peak from matrix components and internal standard in the chromatogram, linearity (r = 0.9993; range 5.0-100.0 mg/L), precision (%RSD 0.7996%), and accuracy (recovery 96.29-101.84%). The results showed that the GC-FID method met all validation parameters according to ICH Q2(R2) and AOAC guidelines. The highest mitragynine content was found in the green variety (1.1321 ± 0.0194 mg/g), followed by red (0.6409 ± 0.0377 mg/g) and white (0.1510 ± 0.0031 mg/g). One-way ANOVA statistical analysis confirmed highly significant differences among kratom leaf varieties (F = 796.136; p < 0.001). The acid-base liquid-liquid fractionation method proved to be 31-71 times more effective than direct extraction. In conclusion, this validated GC-FID method is reliable and applicable for quantitative analysis of mitragynine in kratom product standardization and surveillance.
