Abstract

Kratom (Mitragyna speciosa) merupakan tanaman yang mengandung alkaloid utama mitraginin dengan aktivitas farmakologis menyerupai opioid dan berpotensi menimbulkan risiko kesehatan. Penelitian ini bertujuan untuk memvalidasi metode Gas Chromatography–Flame Ionization Detector (GC–FID) serta menetapkan kadar mitraginin dalam berbagai produk komersial mengandung kratom, meliputi teh kratom, kopi kratom, dan kapsul varietas hijau, putih, dan merah. Validasi metode dilakukan berdasarkan parameter selektivitas, linearitas, presisi, dan akurasi sesuai pedoman ICH Q2(R2). Hasil uji linearitas menunjukkan hubungan linier yang sangat baik antara konsentrasi dan respon dengan koefisien korelasi r ≥ 0,999 dan nilai Vxo sebesar 1,97%. Presisi metode memenuhi kriteria dengan %RSD sebesar 1,041%, sedangkan uji akurasi pada level 80%, 100%, dan 120% menghasilkan % recovery dalam rentang 95,92–100,83%, yang menunjukkan bahwa metode akurat dan presisi. Metode GC–FID yang tervalidasi kemudian diterapkan pada penetapan kadar mitraginin dalam produk komersial. Hasil analisis menunjukkan bahwa teh kratom memiliki kadar mitraginin tertinggi, yaitu 0,2327–0,2846 mg/g dengan rata-rata sekitar 0,7756 mg per kantong teh, diikuti oleh kopi kratom dengan kadar 0,0501–0,0552 mg/g atau rata-rata 0,5335 mg per wadah kopi. Pada sediaan kapsul, kadar mitraginin berturut-turut adalah kapsul varietas hijau sebesar 0,0829–0,1727 mg per kapsul (rata-rata 0,1192 mg), kapsul varietas putih sebesar 0,0873–0,1255 mg per kapsul (rata-rata 0,1019 mg), dan kapsul varietas merah sebagai yang terendah dengan kadar 0,0529–0,0659 mg per kapsul (rata-rata 0,0609 mg). Hasil penelitian ini menunjukkan bahwa metode GC–FID layak digunakan untuk analisis kuantitatif mitraginin dan mengungkap adanya variasi kadar mitraginin yang signifikan pada produk kratom komersial yang beredar.

Kratom (Mitragyna speciosa) contains mitragynine as its major alkaloid, which exhibits opioid-like pharmacological activity and poses potential health risks. This study aimed to validate a Gas Chromatography–Flame Ionization Detector (GC–FID) method and to determine the mitragynine content in various commercial kratom-containing products, including kratom tea, kratom coffee, and capsule formulations of green, white, and red varieties. Method validation was performed in accordance with ICH Q2(R2) guidelines, covering selectivity, linearity, precision, and accuracy. The linearity test demonstrated an excellent proportional relationship between concentration and detector response, with a correlation coefficient (r) ≥ 0.999 and a Vxo value of 1.97%. Method precision met the acceptance criteria, yielding a %RSD of 1.041%, while accuracy evaluated at 80%, 100%, and 120% concentration levels produced percent recoveries ranging from 95.92% to 100.83%, confirming that the method is both accurate and precise. The validated GC–FID method was subsequently applied to the quantification of mitragynine in commercial products. The results revealed that kratom tea contained the highest mitragynine levels, ranging from 0.2327 to 0.2846 mg/g, corresponding to an average of approximately 0.7756 mg per tea bag, followed by kratom coffee, which contained 0.0501–0.0552 mg/g or an average of 0.5335 mg per serving. Among capsule products, the mitragynine content decreased in the following order: green variety capsules (0.0829–0.1727 mg per capsule, mean 0.1192 mg), white variety capsules (0.0873–0.1255 mg per capsule, mean 0.1019 mg), and red variety capsules, which exhibited the lowest content (0.0529–0.0659 mg per capsule, mean 0.0609 mg). These findings indicate that the validated GC–FID method is suitable for the quantitative analysis of mitragynine and highlight substantial variability in mitragynine content among commercially available kratom products.
