Abstract

Infeksi Saluran Kemih (ISK) merupakan salah satu penyakit infeksi yang sering dijumpai di masyarakat dan umumnya disebabkan oleh Escherichia coli. Identifikasi bakteri secara cepat, sensitif, dan spesifik diperlukan untuk mendukung penegakan diagnosis. Salah satu metode yang dapat digunakan adalah Polymerase Chain Reaction (PCR) yang mampu mendeteksi DNA bakteri melalui amplifikasi fragmen DNA target. Penelitian ini bertujuan untuk menganalisis keberadaan DNA Escherichia coli pada sampel urine sewaktu pasien yang diduga mengalami Infeksi Saluran Kemih (ISK) menggunakan metode PCR. Penelitian ini merupakan penelitian deskriptif dengan pendekatan observasional. Sampel penelitian berupa delapan sampel urine sewaktu dari pasien yang diduga mengalami ISK pada periode Januari – Maret 2026 yang diperoleh dari RS TK.III Brawijaya Surabaya. Pemeriksaan dilakukan di Laboratorium Biologi Molekuler Institute of Tropical Disease (ITD) Universitas Airlangga Surabaya pada bulan Januari – Mei 2026. Tahapan penelitian meliputi isolasi DNA, pengukuran kemurnian dan konsentrasi DNA menggunakan spektrofotometer NanoDrop, amplifikasi DNA menggunakan metode PCR dengan primer yang menargetkan gen yaiO, serta visualisasi hasil amplifikasi menggunakan elektroforesis gel agarosa. Hasil visualisasi elektroforesis menunjukkan bahwa tujuh sampel (87,5%) menghasilkan pita DNA berukuran 110 bp yang mengindikasikan keberadaan DNA Escherichia coli, sedangkan satu sampel (12,5%) tidak menunjukkan pita DNA target. Nilai kemurnian DNA berkisar antara 0,37–2,20 dengan konsentrasi DNA sebesar 5,6– 159,9 ng/µL. Berdasarkan hasil penelitian, metode Polymerase Chain Reaction (PCR) mampu mendeteksi keberadaan DNA Escherichia coli pada sebagian besar sampel urine sewaktu pasien yang diduga mengalami Infeksi Saluran Kemih (ISK), sehingga berpotensi menjadi metode penunjang dalam identifikasi bakteri Escherichia coli penyebab Infeksi Saluran Kemih (ISK). Urinary Tract Infection (UTI) is one of the most common infectious diseases encountered in the community and is generally caused by Escherichia coli. Rapid, sensitive, and specific bacterial identification is essential to support the establishment of a diagnosis. One method that can be used is the Polymerase Chain Reaction (PCR), which is capable of detecting bacterial DNA through the amplification of target DNA fragments. This study aimed to analyze the presence of Escherichia coli DNA in random urine samples from patients suspected of having Urinary Tract Infections (UTIs) using the PCR method. This study employed a descriptive research design with an observational approach. The research samples consisted of eight random urine samples collected from patients suspected of having UTIs during the period from January to March 2026, obtained from TK III Brawijaya Hospital, Surabaya. Laboratory examinations were conducted at the Molecular Biology Laboratory, Institute of Tropical Disease (ITD), Universitas Airlangga, Surabaya, from January to May 2026. The research procedures included DNA isolation, measurement of DNA purity and concentration using a NanoDrop spectrophotometer, DNA amplification using the PCR method with primers targeting the yaiO gene, and visualization of the amplification products through agarose gel electrophoresis. The electrophoresis results showed that seven samples (87.5%) produced a 110-bp DNA band, indicating the presence of Escherichia coli DNA, whereas one sample (12.5%) showed no target DNA band. DNA purity values ranged from 0.37 to 2.20, with DNA concentrations ranging from 5.6 to 159.9 ng/µL. Based on these findings, the Polymerase Chain Reaction (PCR) method was able to detect the presence of Escherichia coli DNA in the majority of random urine samples from patients suspected of having UTIs, indicating its potential as a supportive method for identifying Escherichia coli as the causative agent of urinary tract infections.
