Abstract

Latar Belakang: Staphylococcus aureus merupakan salah satu bakteri umum yang
dapat menyebabkan penyakit infeksi yang paling sering di dunia. Seperti infeksi
kulit, endokarditis, pneumonia, osteomielitis, selulitis, impetigo, bahkan hingga
bisa terjadi sepsis. WHO memasukkan S. aureus sebagai bakteri patogen dalam
kategori "high priority" yang memerlukan antibiotik generasi baru. Sehingga
dibutuhkan inovasi lebih lanjut untuk menanggulangi permasalahan dari antibiotik
tersebut. Indonesia merupakan negara yang memiliki aneka ragam tumbuhan herbal
yang dapat diolah dan dimanfaatkan sebagai obat tradisional. Beberapa jenis
tanaman yang tersedia dalam jumlah banyak dan mudah didapat di Indonesia adalah
daun salam dan daun sirih hijau. Daun salam dan daun sirih hijau mengandung zat
aktif seperti flavonoid, tanin, alkaloid dan minyak atsiri sebagai antibakteri.
Kombinasi tanaman yang bersifat antibakteri akan saling mempengaruhi kerja dari
masing-masing senyawa aktif didalamnya, sehingga dapat meningkatkan aktivitas
antibakteri. Oleh karena itu peneliti tertarik untuk melakukan uji aktivitas
antibakteri dengan mengkombinasikan ekstrak daun salam (Syzygium polyanthum)
dan daun sirih hijau (Piper betle Linn) terhadap bakteri S. aureus secara in vitro.
Tujuan: Untuk membuktikan adanya aktivitas antibakteri pada kombinasi ekstrak
daun salam (Syzygium polyanthum) dan ekstrak daun sirih hijau (Piper betle Linn)
terhadap pertumbuhan bakteri Staphylococcus aureus. Metode: Penelitian ini
menggunakan metode kuantitatif True eksperimental laboratorik in vitro dengan
menggunakan desain penelitian Posttest Only Control Group Design. Hasil: Dari
uji analisis menggunakan uji parametrik One Way Anova, Hasil p-value < 0,05
(0,000), ini berarti terdapat perbedaan yang signifikan dari uji kombinasi kombinasi
ekstrak daun salam (Syzygium polyanthum) dan daun sirih hijau (Piper betle Linn)
terhadap bakteri Staphylococcus aureus. Hal ini bermakna bahwa variasi
perbandingan kombinasi ekstrak dan antibiotik signifikan dalam penghambatan
bakteri. Hasil uji Tukey dari penelitian ini diketahui bahwa sebagian besar zona
hambat yang dihasilkan dari uji kombinasi ekstrak dan antibiotik sampel penelitan
menunjukkan perbedaan bermakna. Kesimpulan: Kombinasi ekstrak daun salam
(Syzygium polyanthum) dan daun sirih hijau (Piper betle Linn) memiliki daya
antibakteri terhadap bakteri Staphylococcus aureus.

Background: Staphylococcus aureus is a common bacteria that can cause the most
frequent infectious diseases in the world. Such as skin infections, endocarditis,
pneumonia, osteomyelitis, cellulitis, impetigo, and even sepsis. WHO includes S.
aureus as a pathogeni bacteria in the "high priority" category that requires new
generation antibiotics. So further innovation is needed to overcome the problem of
antibiotics. Indonesia is a country that has a variety of herbal plants that can be
processed and used as traditional medicine. Several types of plants that are
available in large quantities and easy to obtain in Indonesia are bay leaves and
green betel leaves. Bay leaves and green betel leaves contain active substances such
as flavonoids, tannins, alkaloids and essential oils as antibacterials. The
combination of plants that are antibacterial will mutually influence the work of each
active compound in them, thereby increasing antibacterial activity. Therefore,
researchers were interested in testing antibacterial activity by combining bay leaf
(Syzygium polyanthum) and green betel leaf (Piper betle Linn) extracts against S.
aureus bacteria in vitro. Objective: To prove the antibacterial activity of a
combination of bay leaf extract (Syzygium polyanthum) and green betel leaf extract
(Piper betle Linn) against the growth of Staphylococcus aureus bacteria. Methods:
This research uses a true quantitative experimental laboratory method in vitro
using a Posttest Only Control Group Design research design. Results: From the
analysis test using the One Way Anova parametric test, the p-value < 0.05 (0.000),
this means there is a significant difference from the combination test of bay leaf
extract (Syzygium polyanthum) and green betel leaf (Piper betle Linn) on bacteria.
Staphylococcus aureus. This means that variations in the combination of extracts
and antibiotics are significant in inhibiting bacteria. The results of the Tukey test
from this study showed that most of the inhibition zones resulting from the
combination test of extracts and antibiotics on research samples showed significant
differences. Conclusion: The combination of bay leaf extract (Syzygium
polyanthum) and green betel leaf (Piper betle Linn) has antibacterial power against
Staphylococcus aureus bacteria.
